Kinetics

Neben strukturellen und thermodynamischen Eigenschaften von Protein-Interaktionen untersuchen wir auch die Dynamik der einzelnen Wechselwirkungen, um eine vollständige Charakterisierung zu erreichen. Mit Hilfe der stopped-flow Technik messen wir die Geschwindigkeiten der Assoziations- und Dissoziationsprozesse der Komplexe von Proteinen miteinander oder mit Co-Faktoren. Zur Detektion verwenden wir vor allem die Änderung der Fluoreszenz-Intensität oder -Polarisation. Um kompliziertere Reaktionsmechanismen, wie die einzelnen Schritte der enzymkatalysierten Hydrolyse von GTP, aufzuklären, verwenden wir die rapid quench flow Methode. Diese erlaubt nicht nur eine zeitlich hoch aufgelöste Bestimmung aller Reaktanden- und Produktkonzentrationen, sondern ermöglicht auch die Beobachtung von kurzzeitig auftretenden Reaktionsintermediaten.

Stopped-Flow Measurements



T-Jump

A temperature-jump instrument allows for resolution of fast protein folding and complex formation processes on the microsecond to millisecond time scale. This method enables us to study the impact on protein folding and binding evoked by co-solutes like salts and ionic liquid compounds and by macromolecular crowding systems like dextran and polyethylene glycol1,2. In particular, we can investigate fast kinetics of fundamental biochemical processes which is rarely addressed in protein solvation studies. Though, relaxation kinetics give detailed insight into changes of the molecular mechanism of protein folding and complex formation, respectively, as a consequence of variations of the solvent environment.

The temperature-jump is a pertubation method enabling us to measure fast relaxation kinetics. The rapid heating of the solution under investigation is achieved by an electrical current generated by discharging a large capacitance within 10 ms. The following chemical relaxation of the biochemical system adapting to the new equilibrium value is monitored by changes of optical density and of fluorescence, respectively.
