Synaptic plasticity in the central nervous system entails a wide variety of mechanisms, encompassing both chemical and electrical synapses. Electrical synapses (gap junctions of the nervous system) are a key component of the neural circuitry throughout the CNS. This is most apparent in the vertebrate retina or in the hippocampus, where gap junctions play critical roles in neural processing. In the retina these include reducing noise at photoreceptor synapses, establishing receptive field sizes of many neurons, coordinating firing of spiking neurons, and establishing oscillations in neural networks. These functions are closely controlled during light and/or dark adaptation, influencing the sensitivity and resolution of the retina, and accounting for a large part of the network plasticity. In the hippocampus (and in other regions like the cortex or thalamus as well) electrical synapses help synchronize the activity of brain cells by regulating specific brain rhythms. These oscillations are, in a basic sense, associated with the way how the brain regulates behavioural states.
 The major focus of my work is dedicated to the molecular and cellular mechanisms that control network properties mediated by electrical synapses in the retina. Our work in this area includes the characterization of two neuron-specific gap junction proteins (connexin 52.6, Cx55.5), which are exclusively expressed in horizontal cells of the zebrafish. Other connexins (Cx) and members of a recently discovered novel family of gap junction termed pannexins (Panx) have been cloned from the zebrafish and the expression localized in the retina and brain. Having completed the cloning and expression analysis we are now examining how specific cellular pathways modulate the functions of these gap junction channels, and how light/dark adaptation drives these pathways.
 A second focus is related to studies of the function of the neuron-specific connexins and pannexins in the rodent hippocampus and cortex. In these studies we are interested in the molecular networks associated with Cx36 and Panx1/Panx2. Having identified distinct groups of Cx and Panx interacting proteins and regulated genes we are at the beginning to uncover the signalling pathways related to activities of electrical synapses in the rodent brain.
 My research is based on a broad-based expertise in cellular and molecular neuroscience and in imaging. Ongoing research projects will include in vitro molecular, cellular and biochemical studies, imaging analysis using in particular confocal microscopy or live cell imaging, and transgenic zebrafish technologies which we currently establish in the laboratory. I do collaborate intensively with laboratories with expertises in functional genome and proteome analysis or electrophysiology.
